Polyacetylenes from the florets of Carthamus tinctorius and their cytotoxicity

Two new polyacetylenes, (2R, 11S, 12S)-(3E)-tridecene-5,7,9-triyne-1,2,11,12-tetraol (1) and (8S)-deca-4,6-diyne-1,8-diol-8-O-beta-D-glucopyranoside (2), together with six known polyacetylenes (3-8), were isolated from the florets of Carthamus tinctorius. Their structures were elucidated on the basis of spectroscopic data (NMR and HRMS) and Mo2(OAc) 4-induced circular dichroism. Compounds 4 and 5 were cytotoxic to the human cancer cell lines HL-60 (IC50=6.40, 3.66 mu M), THP-1 (IC50=8.67, 6.69 mu M) and PC-3 (IC50=11.30, 10.92 mu M), comparable to the positive control, fluorouracil (IC50=2.58, 3.95 and 22.35 mu M, respectively).
