Abstract新疆雪莲 (Saussurea involucrata Kar et Kir)系菊科风毛菊属植物,主产于新疆天山、昆仑山,生长在雪线以上,民间以全草入药,具有活血通经、散寒除湿、强筋助阳等功效,因此,被用来作为一种药物主要用于治疗风湿性关节炎、妇科疾病。现代药理研究表明,其具有清除自由基、抗疲劳、抗癌、抗炎和免疫调节作用。目前,雪莲全草的化学成分已经研究透彻,但雪莲种子成分的研究很少。 我们采用GC-MS、ICP元素分析法和常规分离方法对雪莲种子提取物进行化合物分离鉴定出牛蒡子苷、棕榈酸、亚油酸、油酸、花生四烯酸等多种化合物。此外还从雪莲种子检测到无机元素21种,其中人体必需常量元素6种,微量元素12种。 对雪莲种子脂肪酸含量进行测定得出其饱和脂肪酸含量为5.47 %,不饱和脂肪酸含量为94.07 %,其中含有人体必需脂肪酸亚油酸(73.95 %)和花生四烯酸(0.11%)。对雪莲种子挥发油进行测定时,检定出烷、烯、醇、酚、醛、酮、酸、酯类等69种化合物,占总量的81.19 %。对雪莲种子分级提取物进行抗氧化活性测定时发现其提取物均有活性,氯仿提取物活性最好。 采用LC-MS联用技术对雪莲种子、苞叶和叶片的化学成分对比分析。结果表明,三者之间化学成分差异较大,但叶片和花中类似化学成分较多。建立HPLC 法测定雪莲种子提取物中牛蒡子苷含量的方法,为新疆雪莲特征性化合物的指认提供科学依据。所建立的方法简便、准确、重现性好,并具有良好稳定性,为准确测定雪莲种子中牛蒡子苷含量提供可靠依据。
Other AbstractSaussurea involucrata Kar. et Kir. (family Asteraceae)is one of the precious and endangered medicinal herbs in traditional Uighur medicine.It is distributed in Tianshan Mountains and Kunlun Mountains area and grows above the snow line. According to the theories of traditional Chinese medicine, it has the efficiency of warming kidney and activating ‘yang’, expelling wind and eliminating dampness, including menstruation and promoting blood circulation. Therefore, it has been used as a principal drug for the cure of rheumatic arthritis and gynecological disorders. Modern pharmacological studies have demonstrated that it possesses activities such as free radical scavenging, antifatigue, anti-inflammation, anticancer, and immu- nomodulation. Chemical constituents from S. involucrata has been researched thoroughly, but components in seeds of S. involucratain are researched rarely. Recently, GC-MS, ICP elements analysis method, and conventional method for isolation have been used to analyze fatty acid, inorganic elements and chemical constituents in seeds of S. involucrata. Finally, we get arctiin, sixteen acid, linoleic acid, oleic acid, β-sitosterol from S. involucratain seeds. In addition, we get 21 elements ,including 6 essential macro elements, 12 essential trace elements. The result of fatty acid content determination of S. involucratain seed showed the content of saturated fatty acid and unsaturated fatty acid are 5.47 %, 94.07 %, respectively. The essential fatty acids linoleic acid (73.95%) and arachidonic acid (0.11%) have been determined. Alkane, alkene, alcohols, phenols, aldehydes, ketones, acids, esters and other kinds 100 compounds have been determined in the volatile oil of S. involucrata seed. 69 of them have been identified and the conent of is 81.19% of the total. Antioxidant activity of S. involucrata seed grading extract have been tested. And all the extracts showed activity, while chloroform extracts activity is the best. LC-MS technology has been used to analyze chemical compositions in seeds, flowers and inflorescence of S. involucrata. The results showed that, among the three chemical composition difference is notable, but the inflorescence and flowers are more similar chemical composition. To establish HPLC method for the determination of arctiin in seeds of S. involucrata, and provide the scientific basis for the identification of characteristic compound of S. involucrata. This method is simple, accurate, and has good stability, while provide a reliable basis for the accurate determination of arctiin in seeds of S. involucrata.
