摘要

独尾草属植物(百合科)主要分布在亚洲的中部以及西部,被称为狐尾百合或沙漠蜡烛。独尾草属植物大约有45种,中国分布4种。阿尔泰独尾草(Eremurus altaicus(Pall.) Stev.)是百合科独尾草属植物的一种,在我国仅仅分布在新疆的天山和阿尔泰山。独尾草属植物在中亚各国已有悠久的药用以及食用历史,而阿尔泰独尾草的化学成分与生物活性还未见报道。在阿尔泰独尾草的化学成分研究方面,利用硅胶柱色谱,凝胶柱色谱,制备液相色谱等方法首次从阿尔泰独尾草全草中分离得到11个单体化合物,并通过红外,紫外光谱,核磁,质谱等确定其结构。大部分为醌类化合物,其中有新的对应异构体萘酮类化合物首次从自然界中发现,利用手性色谱柱进行拆分,得到一对对映异构体,并利用电子圆二色谱分别确定其绝对构型。在阿尔泰独尾草的生物活性方面,经过活性筛选,发现阿尔泰独尾草乙腈部位具有很好的抗氧化和PTP1B抑制活性,并以乙酸乙酯和乙腈部位的活性尤为突出,进一步筛选初步得到结论,阿尔泰独尾草活性突出的组分集中在中小极性分子上。由于阿尔泰独尾草具有良好的PTP1B抑制活性,所以对活性成分的提取纯化等不同工艺条件进行了探究。由单因素-正交实验得到最佳提取工艺。然后对十种大孔树脂进行了静态吸附-解吸附的筛选,并对筛选的最佳型号树脂HPD-300的不同极性溶剂的洗脱组分进行了活性测定,最终得到了阿尔泰独尾草属植物的有效降糖部位的制备方法。阿尔泰独尾草中降糖活性成分提取的最佳工艺为70%乙醇溶液,料液比1:30,提取时间2小时,提取温度70℃。并通过HPD300型号的大孔树脂进行纯化。通过对阿尔泰独尾草的化学成分研究,我们对阿尔泰独尾草有了一个初步的认识,而阿尔泰独尾草出色的降糖活性还有待进一步研究,对于弄清楚是什么物质起到了生物活性是今后的工作重点。

The genus Eremurus (Liliaceae), mainly distributed in the western and central Asia, is known as foxtail lilies or desert candles. It comprises 45 speices, and four of them have been found in the People’s Republic of China。Eremurus altaicus (Pall.) Stev. belongs to the Eremurus genus of the lily family, which restrictively distributed in the dry area of the Mountain Altai and Tianshan Mountain in Xinjiang Uighur Autonomous Region. The Eremurus genus has a long history of medicinal and edible in the Central Asian countries.And the chemical composition and biological activity of the Eremurus altaicus (Pall.) Stev. has not been reported till now. In the chemical composition of the Eremurus altaicus (Pall.) Stev., 11 compounds were isolated from the whole plant by different means, for instance, silica gel column , Sephadex LH-20 and preparation HPLC. Their structures were identified by MS and NMR spectral data. The majority of the compounds were Quinones compounds. A new racemic 1,2,3,4-tetrahydro-1-naphthalenone was first isolated from nature .Then purified by chiral HPLC to yield a pair of enantiomers. The absolute configurations of the pair of enantiomers were elucidated by quantum chemical ECD calculations. In terms of the test of biological activity, the Eremurus altaicus (Pall.) Stev. extraction showed good antioxidant activity and PTP1B inhibitory activity. Especially, the ethyl acetate and acetonitrile extraction exhibited outstanding biological activity. After the further evaluating, we draw a preliminary conclusion that the excellent biological activity may focus on the medium and small polar molecule parts of the Eremurus altaicus (Pall.) Stev. Because of the excellent PTP1B inhibitory activity, different process conditions of the extraction and purification of active ingredients were explored. The best extraction process was obtained by the single factor-orthogonal experiment. Ten kinds of macroporous resin Static adsorption–desorption were then tested.We evaluated the biological activity of eluted fractions of different solvents through the HPD-300 macroporous resin which was the best model.At last we got the best approach to Eremurus altaicus (Pall.) Stev. effective hypoglycemic site. Through the study of the chemical composition of Eremurus altaicus (Pall.) Stev., we have a preliminary understanding of the plant, the excellent hypoglycemic activity of Eremurus altaicus (Pall.) Stev. remains to be further research to figure out what the substance played a biological activity in the further work. Key word: Eremurus altaicus (Pall.)
